Cytonics Concludes Phase 1 Trials for Osteoarthritis Drug CYT-108

Jupiter-based biopharmaceutical firm Cytonics Corporation has finalized the clinical study report for its first-in-human trial of CYT-108, a recombinant protein therapy designed to treat knee osteoarthritis. The milestone transitions the company toward upcoming Phase 1b/2a development stages and broader regulatory engagement for its lead biologic candidate.

The randomized, double-blind study evaluated the safety and exploratory efficacy of CYT-108 across multiple Australian clinical sites. Twenty-two patients with mild-to-moderate primary osteoarthritis received two intra-articular injections spaced 90 days apart, with researchers tracking outcomes over a six-month period. Company leadership views the completion of this report as a foundational step in validating the drug’s potential as a disease-modifying therapy.

Unlike conventional treatments that target isolated molecular pathways, CYT-108 is engineered as a recombinant variant of Alpha-2-Macroglobulin, a broad-spectrum protease inhibitor. By addressing multiple protease pathways simultaneously, the therapy aims to mitigate cartilage degradation more effectively than existing small-molecule inhibitors. Cytonics draws confidence from its previous success with the APIC system, an FDA-cleared device that utilizes autologous A2M, providing a clinical roadmap for the safety of high-concentration A2M delivery in arthritic joints.
