Kelun-Biotech's ADC Combo Hits Primary Endpoint in Lung Cancer Trial

A Phase III study has demonstrated that the antibody-drug conjugate sacituzumab tirumotecan, when paired with pembrolizumab, significantly improves progression-free survival in patients with PD-L1-negative non-squamous non-small cell lung cancer, marking a potential shift in the first-line treatment landscape for those with limited immune responses.

The OptiTROP-Lung06 trial, conducted by Sichuan Kelun-Biotech, achieved its primary endpoint by replacing traditional pemetrexed and platinum-based chemotherapy with the TROP2-directed ADC sac-TMT. By combining the ADC with an immune checkpoint inhibitor, researchers observed not only a statistically significant improvement in progression-free survival but also a positive trend in overall survival. This mechanism is designed to activate the immune microenvironment, specifically targeting tumors that typically show poor response to standard immunotherapy.

Professor Caicun Zhou of Shanghai East Hospital noted that while current standard-of-care combinations have improved outcomes, long-term survival remains a challenge for this patient population. The trial results suggest that this combination therapy offers a viable alternative to existing protocols. Kelun-Biotech, which has seen its drug sac-TMT earn multiple breakthrough designations in China, plans to initiate discussions with the National Medical Products Administration regarding these findings. The company currently manages ten registrational studies for the drug across various cancer types, signaling a broader push to integrate the ADC into standard clinical practice.
