By applying six distinct aging models—including ProtAge, OrganAge, and PAC—the team observed a consistent trend of biological age reversal in the treatment group, with some metrics indicating a shift of up to six years. The study demonstrates that the drug's impact on aging biomarkers appears partially independent of its respiratory benefits in lung function.
Insilico Medicine, the developer of the TNIK-targeting molecule, utilized its generative chemistry platform to identify the target and design the candidate in roughly 18 months. Beyond the clinical results, the company established a standardized blueprint for embedding geroscience endpoints into traditional drug trials. Nobel laureate Michael Levitt noted that the consensus across diverse models, which share neither training data nor features, adds significant weight to the findings. While the current data cannot fully decouple the drug's anti-fibrotic activity from its systemic anti-aging effects, the company is now advancing rentosertib into Phase III trials.
