The findings represent a 6.4-month improvement in median overall survival compared to the 27.9 months observed in the chemotherapy-only arm. While the raw hazard ratio of 0.87 did not reach statistical significance, researchers noted that the trial design allowed 76% of patients in the control group to cross over to amivantamab monotherapy following disease progression. When adjusted for this crossover using an inverse probability of censoring weighting model, the survival benefit became statistically significant with a hazard ratio of 0.57.

Dr. Chul Kim of the Georgetown Cancer Institute, who presented the data, described the regimen as a potential new foundation for first-line treatment in this patient population. The PAPILLON study, which enrolled 308 participants, also reported that the safety profile for the intravenous combination remained consistent with historical data. Beyond survival metrics, the treatment demonstrated positive patient-reported outcomes, specifically regarding the time to worsening of cancer-related symptoms.
