Alpheus Medical Launches Clinical Trial for Butterfly Glioblastoma

Patients diagnosed with butterfly glioblastoma, an aggressive brain cancer crossing both cerebral hemispheres, have a new clinical path forward. Alpheus Medical has initiated the Butterfly trial to test its PoMA therapy, a drug-led treatment using ultrasound, which requires enrollment before the start of standard chemoradiation protocols.

The study, registered as NCT07862647, aims to address a patient population frequently excluded from traditional glioblastoma trials due to the tumor's diffuse, bilateral spread. With a median survival rate of just three to six months, butterfly glioblastoma often renders surgical resection impossible. Dr. David Reardon of the Dana-Farber Cancer Institute noted that the condition’s distribution demands a departure from standard oncology practices, highlighting the need for targeted strategies that span both hemispheres.

Alpheus Medical’s PoMA therapy involves the oral administration of 5-aminolevulinic acid, which accumulates in tumor cells. Once localized, the substance is activated by low-intensity defocused ultrasound while the patient remains awake in an outpatient setting. The Dent Neurologic Institute in Buffalo, New York, is currently the active site, with plans to expand to additional U.S. centers.
