Roche bets on Rona Therapeutics' ALK7 RNAi for obesity treatment

Roche has secured an exclusive global license for Rona Therapeutics’ experimental RNAi candidate targeting ALK7, a protein linked to fat distribution. The deal, which focuses on a novel approach to adipocyte biology, could bring Rona up to $1.16 billion in upfront payments and future development milestones.

The partnership centers on Rona’s proprietary extrahepatic delivery platform, designed to silence the ACVR1C gene, which encodes ALK7. While current obesity treatments like incretins primarily target appetite through the central nervous system, this RNAi therapy aims to act directly on adipose tissue. By inhibiting ALK7, the treatment seeks to reduce visceral fat depots while preserving lean muscle mass, potentially offering a complementary mechanism to existing weight-loss drugs.

Stella Shi, CEO and Founder of Rona Therapeutics, described ALK7 as a compelling target for addressing the durability of weight loss. For Roche, the agreement bolsters its existing cardiovascular, renal, and metabolism pipeline. Under the financial terms, Rona is set to receive $100 million in combined upfront and near-term payments, with additional tiered royalties tied to future global net sales. The candidate remains in the investigational stage, with development and commercialization responsibilities shifting to Roche as they aim to expand their footprint in metabolic health.
