BioLineRx and Hemispherian report synergy in ovarian cancer trial

New preclinical data shows that combining the experimental drug GLIX1 with PARP inhibitors triggers synthetic lethality in ovarian cancer models. The findings suggest a potential breakthrough for patients with homologous recombination-proficient tumors, a group that typically sees limited benefit from current PARP inhibitor therapies.

The study utilized patient-derived xenograft models to test the efficacy of GLIX1 alongside the PARP inhibitor olaparib. Results revealed that low-dose combinations outperformed both the control group and individual monotherapies. Notably, the synergistic effect produced tumor reduction comparable to cisplatin, the current standard-of-care chemotherapy.

BioLineRx Chief Executive Officer Philip Serlin noted that the trial confirms earlier in vitro observations, providing a clear path to address platinum resistance. Driven by these results, the companies intend to incorporate an ovarian cancer cohort into the expansion phase of their ongoing Phase 1/2a clinical trial. This move aims to validate whether GLIX1 can successfully sensitize HR-proficient tumors—which account for roughly half of high-grade serous ovarian cancers—to existing PARP-targeted treatments.
